Axonal transport of membranous and nonmembranous cargoes: a unified perspective

نویسنده

چکیده

Membranous and nonmembranous cargoes are transported along axons in the fast and slow components of axonal transport, respectively. Recent observations on the movement of cytoskeletal polymers in axons suggest that slow axonal transport is generated by fast motors and that the slow rate is due to rapid movements interrupted by prolonged pauses. This supports a unified perspective for fast and slow axonal transport based on rapid movements of diverse cargo structures that differ in the proportion of the time that they spend moving. A Flash feature (http:// www.jcb.org/cgi/content/full/jcb.200212017/DC1) accompanies this Mini-Review.

Viral regulation of the long distance axonal transport of herpes simplex virus nucleocapsid.

Many membranous organelles and protein complexes are normally transported anterograde within axons to the presynaptic terminal, and details of the motors, adaptors and cargoes have received significant attention. Much less is known about the transport in neurons of non-membrane bound particles, such as mRNAs and their associated proteins. We propose that herpes simplex virus type 1 (HSV) can be...

The slow axonal transport of alpha-synuclein--mechanistic commonalities amongst diverse cytosolic cargoes.

Slow axonal transport conveys perikaryally-synthesized cytosolic proteins in a rate-class called Slow Component-b (SCb). One such protein--α-synuclein--is largely conveyed in SCb, and is also a key player in a group of neurodegenerative diseases called synucleinopathies. Axonal transport defects of α-synuclein have been hypothesized to play a role in synucleinopathies, but mechanisms moving α-s...

Dynactin

What does it do? It’s part of the molecular machinery that drives a whole host of movements within the cell. Almost every organelle that moves towards the slow-growing ends of microtubules uses cytoplasmic dynein, and so also needs dynactin. Cargoes include the Golgi apparatus, ER–Golgi traffic, most endocytic organelles, nuclei and material for retrograde axonal transport. More unusual cargoes...

Seeing the unseen: the hidden world of slow axonal transport.

Axonal transport is the lifeline of axons and synapses. After synthesis in neuronal cell bodies, proteins are conveyed into axons in two distinct rate classes-fast and slow axonal transport. Whereas fast transport delivers vesicular cargoes, slow transport carries cytoskeletal and cytosolic (or soluble) proteins that have critical roles in neuronal structure and function. Although significant p...
